I have been chasing a vibration in my T100 for years now, which although not the same truck the same principles apply. If its only in first gear I would say its inside the trans itself.

However, it could be any of the following:
-Bent cv axle
-bent front axle
-bad rear wheel shafts
-Balance issues with tires/wheels
-Body mounts being to loose or tweaked
-leaf springs/leaf spring bushings bad
-rear axle bent
-bent spindles
-bad wheel bearings
-drive shaft issues
-u joint issues
-center driveshaft bearing (if equipped)
-hanging brake calipers/wheel cylinders
-etc the list can go on and on

I would suggest you either put it up on a lift and put it into gear or on some stands and have someone drive it while in the air, might be able to see whats moving or spinning with a shake to it. I would check all the above I mentioned, may also want to remove the rear driveshaft and put it into 4wd and see if you feel it then, if you dont you know its in the rear.

In my case I have concluded the front of my frame is slightly tweaked or the body is. I also gave up and in the process of selling my truck to someone who doesnt care if it vibrates. Ill tell you right now, if you cant figure it out get rid of it if it annoys you-I shoulda done that long ago instead of spending tons of cash fixing.

Im willing to bet its your tires starting to spin. Full throttle in first gear with a v8 is gonna spin them a little. That would also explain why it doesnt do it in 4wd. It definatley isnt a trans issue if it doesnt do it in 4wd. Anyway, I really wouldnt worry about it, how often are you taking off full throttle? I'm a pretty hard driver, but its pretty rare that im wide open in 1st gear.

Ah yes could be rear axle hop due to the spin of the tires. Some better springs will help that greatly.

I have the same vibration, only in 1st gear, and it only does it if I floor it and it shifts from 2nd down to first. Just for the heck of it,, I tried it when I was pulling my 3000lb trailer and it was worse. I assumed it's ujoints or something where as when I put the weight of the trailer on the truck it gets worse. I just greased all the joints last night so we will see if that helps out.

I also pull a 5000 lb trailer and mine also gets worse pulling the trailer .It seams like the drive shaft to me.The center support in the rubber seams allful slopy to me and i think that could cause it by letting the drive shaft kick to side some.

THat center support bearing should have a little play in it, but not much. They are pretty cheap to replace, like 50 bucks or so.

Still, the springs could be wrapping if the bushings are worn or you have alot of torque-which is evident. Need traction bars if thats the case.
