I also jacked up the truck and spun the tire and one went forward and the other went in reverse, I know that it means that its an open diff but could I have limited slip and the clutch packs in it are just worn out. Like I said I bought the truck used with 58,000 miles on it and it now has 66,000 miles and I don't know if the previous owner ever had the diff serviced, because I bought it at a Lexus dealer.

Unless your truck is a very late production '01 it is highly unlikely to have a factory LSD as they were not generally available as an option until the 2002 model year. Possible to have aftermarket Kazuma or other brand as they are avaulable for the '01.
