I noticed lately that my transmission runns rough between 40 and 45 mph. I notice this mostly when going on a slight incline. I have nearly 20000 miles on the car. Any suggestions?

Sounds like you have an I4, in 5th gear and the engine is lugging at 1,400-1,500 RPM.
Give it some gas so it shifts down or shift down into 4th until you get over 50-55 mph.

I have a 2006 camary with a 5 speed automatic I always just take it out of overdrive in town until I hit the highway then I put it back into overdrive this seems to end the lugging the engine in town traffic and I was told by a good transmission person the trans will last longer this way.
