No, it's not possible without significant structural modification. It's easy to lift trucks because they have body on frame construction. The Highlander has a Uni-body, like a car, so there is no way to "lift" the body off the frame, as they are the same thing.
Also, you can't really lift the suspension, because it's geometry doesn't allow. It was based off the Camry, and this wasn't a design consideration. Honestly, the HL won't be able to do most of the things you would lift a truck for anyway.
You can put on slightly larger tires....
