No, there are standard air valves on an Indirect TPMS system as used on the Highlander. (The 2008 HL now does get the Direct type TPMS with a sensor valve on each of the 5 tiresas found on the Tundra and Land Cruiser).

This Indirect system use the ABS speed sensors (4 ), one at each wheel to determine rotational speed differences among the four rolling tires(not the spare for obvious reasons. Any difference at one or more tires indicates a tire pressure problem.
