If someone can answer this question, I would really appreciate it. Would a TRD Off-Road Package on a V6 Tacoma affect gas mileage?

I can't address a Tacoma directly but on a Tundra the TRD package is just different shocks and a stiffer suspension, bigger wheel lip moldings, the MPG is the same for all models with the V-8 regardless if it has the TRD package.

The only thing depending on year would be the bigger tires would might affect MPG a little bit, 31x10.5's on my '98 seemed to net a little worse then my '01 did for mpg, then again changes in the ecu occured over that timespan too. But if it were anything dramatic it (should be) listed @ fueleconomy.gov as a different model. But the diff/susp/etc. shouldn't really amount to any noticeable mileage change.

A set of shocks,locking rear is about it.you can get the same size tire on one without the off road package.
